General Notes
This product (Catalog No. DPABH-10662) is a rabbit-derived IgG polyclonal antibody raised against human GPR22. This antibody exhibits cross-reactivity with GPR22 from mouse and rat. It has been affinity purified by protein A. GPR22, or G protein-coupled receptor 22, is an orphan receptor protein encoded by the GPR22 gene on human chromosome 7q22. It belongs to the class A (rhodopsin-like) family of G-protein coupled receptors (GPCRs) and is characterized by seven transmembrane domains typical of this superfamily. As an orphan receptor, its endogenous ligand remains unidentified, distinguishing it from ligand-bound GPCRs. GPR22 primarily signals through the inhibitory G(i)/G(o) protein pathway, modulating cellular responses such as adenylate cyclase inhibition. Functional studies indicate its involvement in ciliogenesis, where it regulates primary cilia length in cells, potentially influencing sensory and signaling processes. In cardiomyocytes, GPR22 overexpression activates the protective Akt signaling pathway while suppressing ERK activation, thereby reducing ischemia-induced myocardial injury in experimental models. The receptor exhibits a restricted expression pattern, with high levels in the brain and heart across humans and rodents, and lower expression in other tissues such as the kidney and testis. Genome-wide association studies (GWAS) have linked genetic variants near GPR22 to susceptibility for knee osteoarthritis, suggesting a role in chondrocyte-like cells and cartilage homeostasis. Additionally, GPR22 variants have been associated with abdominal obesity-metabolic syndrome in some genetic studies.
DPABH-10662 is suitable for use in Western blot (WB) (1:500–1:2,000), IF (immunofluorescence) (1:50–1:200) applications.
DPABH-10662 is supplied as a liquid formulation in PBS with 0.02% sodium azide, 50% glycerol, pH7.3. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
