General Notes
This product (Catalog No. DPABH-20357) is a rabbit-derived polyclonal antibody raised against human CEP85L. It has been affinity purified by Protein A. CEP85L is a protein-coding gene located on chromosome 6q22.31 that encodes centrosomal protein 85-like (CEP85L), a 805-amino acid protein characterized by a C-terminal coiled-coil domain and localization to the pericentriolar material of centrosomes. This protein plays an essential role in centrosomal organization and is critical for neuronal cell migration during brain development, with knockdown studies in mice demonstrating defects in cortical neuron positioning. Additionally, CEP85L was initially identified as a serologically defined breast cancer antigen known as NY-BR-15, though its precise oncogenic mechanisms remain under investigation. Heterozygous pathogenic variants in CEP85L, including missense mutations in a conserved N-terminal region, splice site alterations, and disruptions to the translation initiation codon, cause lissencephaly 10 (LIS10), a neurodevelopmental disorder characterized by abnormal brain folding, intellectual disability, and epilepsy, often presenting with de novo or inherited autosomal dominant transmission. These variants likely exert a dominant-negative effect rather than simple haploinsufficiency, as evidenced by reduced protein levels and disrupted centrosomal function in patient-derived models. Beyond neurology, emerging evidence links CEP85L fusions or dysregulation to oncogenesis, including roles in breast and other cancers, highlighting its multifunctional significance in cellular processes.
DPABH-20357 is suitable for use in IF/ICC 1 (100-1:500) application.
