General Notes

This product (Catalog No. DPABH-19433) is a rabbit-derived IgG polyclonal antibody raised against human FCGR1A, targeting the amino acid region 101–200 of the protein. This antibody exhibits cross-reactivity with FCGR1A from mouse and rat. It has been affinity purified by protein A. This gene encodes a protein that plays an important role in the immune response. This protein is a high-affinity Fc-gamma receptor. The gene is one of three related gene family members located on chromosome 1.
DPABH-19433 is suitable for use in Western blot (WB) (1:500–1:2,000), ICC/IF (1:50–1:200), ELISA (1:5,000–1:10,000) applications.
DPABH-19433 is supplied as a liquid formulation in 0.01M TBS (pH7.4) with 1% BSA, 0.02% Proclin300 and 50% Glycerol. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.

Effect of scopoletin on phagocytic activity of U937-derived human macrophages: Insights from transcriptomic analysis

Abstract

Scopoletin is a botanical coumarin. Notably, scopoletin effect on phagocytic activity has not been addressed on transcriptomic level. Accordingly, this study investigated the effect of scopoletin on phagocytosis-linked gene transcription. Whole phagocytosis transcriptional profiling of stimulated U937-derived macrophages (SUDMs) in response to scopoletin as compared to non-treated SUDMs was studied. Regarding scopoletin effect on 92 phagocytosis-linked genes, 12 of them were significantly affected (p-value < .05). Seven genes were down-regulated (CDC42, FCGR1A/FCGR1C, ITGA9, ITGB3, PLCE1, RHOD & RND3) and five were upregulated (DIRAS3, ITGA1, PIK3CA, PIK3R3 & PLCD1). Moreover, scopoletin enhanced phagocytic activity of SUDMs. The current results highlighted the potential use of scopoletin as immunity booster and as an adjuvant remedy in management of some autoimmune reactions. To the best of our knowledge, this is the first report that unravels the effect of scopoletin on phagocytosis via transcriptomic analysis.

Abstract

IgG FcRs are important mediators of immunity and play a key role during Ab-based immunotherapy. Within the leukocyte IgG receptor family, only Fc gamma RI is capable of IgG binding with high affinity. Fc gamma RI exists as a complex of a ligand binding a-chain and an FcR gamma-chain. The receptors' alpha-chain can, furthermore, elicit several functions independent of the ITAM-bearing FcR gamma-chain. Functional implications of high-affinity IgG binding and mechanisms underlying FcR gamma-chain-independent signaling remain unclear to this day. In this paper, we provide an overview of past literature on Fc gamma RI and address the implications of recently described interactions between cytosolic proteins and the Fc gamma RI alpha-chain, as well as cytokine-enhanced Fc gamma RI immune complex binding. Furthermore, an analysis of potential polymorphisms within the FCGR1A gene is provided. The Journal of Immunology, 2011, 186: 2699-2704.
