Evidence of a role of ANGPTL6 in resting metabolic rate and its potential application in treatment of obesity

Abstract
Aim. ANGPTL6 (Angiopoietin-related growth factor 6) is a circulating protein which is suggested to antagonize obesity. The purpose of this study was to evaluate a potential relationship between fasting serum ANGPTL6 and resting metabolic rate (RMR) as well as the body composition in obese and subjects with normal weight. Methods. Participants were 62 obese and 41 non-obese subjects who were assessed following an overnight fasting for RMR by means of indirect calorimetry. Body composition was measured using Bodystat devise. Serum ANGPTL6 levels were quantified by ELISA method. Results. Based on ROC analysis best RMR/kg cut-off value for predicting the risk of obesity was 20 kcal/24h /kg. The participants with RMR/kg >= 20 kcal/24h/kg were considered as and subjects with RMR/kg<20 kcal/24h/kg were categorized as. In group I, 72.3% of subjects were obese, whereas, 47.4% subjects in group II were suffering from the disease. Participants in group II who showed significantly lower HDL and ANGPTL6 levels. Moreover, we found significantly higher triglyceride and hs-CRP levels in this group. There was significant difference in weight, body mass index, fat mass, visceral fat, RMR/kg, fasting serum glucose, insulin and hs-CRP among those with different levels of the serum ANGPTL6 concentration. We found higher values of RMR/kg in subjects with higher circulating ANGPTL6 concentration. Conclusion. ANGPTL6 affects RMR and significantly improves lipid profile and slightly does so regarding insulin concentrations and sensitivity to it. Further study is warranted as it seems that the results of this study might potentially lead to advent of a pharmacological treatment for obesity.
Bovine Angptl6 gene effects on growth traits in three Chinese native cattle breeds

Abstract
As a circulating orphan peptide secreted by liver, Angptl6 gene might have great effects on growth and development of animals. The objective of this study was to explore sequence variants (SVs) of Angptl6 and to analyze their associations with growth traits in 737 individuals belonging to 3 Chinese native cattle. Herein, 3 variants were revealed by DNA pools sequencing and PCR-RFLP (or ACRS-PCR-RFLP) methods. These SVs were located at nt 2359, nt 2403 and nt 3258 (viz. SV1, SV2, SV3) respectively. All the analyzed populations were at Hardy-Weinberg equilibrium by chi(2)-test. The linkage disequilibrium (LD) and haplotype analyses of 3 SVs revealed 8 different haplotypes. Thereinto, 2 dominant haplotypes (TCG and CCG) occurred with a frequency of 63.17% and 18.67% respectively. Association analysis showed the SV1 was significantly associated with BLI-6, and the SV2 and SV3 were significantly associated with CGI-24 and BHI-6 of individuals of Nanyang breed respectively. Further, combined SVs analysis revealed the ADG-18 of individuals with diplotype TC-CC-GG was higher than other combined genotypes in SV1-SV2-SV3 locus. Moreover, CG-18 with TC-GG and ADG-18 with TT-GT were preponderant in SV1-SV3 locus. Thus, 3 SVs showed great comprehensive effects on growth traits of Nanyang cattle, which indicated that they could be contributed to genetic and breeding programmes in cattle. Although there was no sufficient evidence these SVs greatly effect on the growth traits of cattle, the significant effects of these variants with these parameters could not be ignored.
