ANGPTL6 Introduction
ANGPTL6, also known as angiopoietin-like protein 6, is a secreted glycoprotein that belongs to the angiopoietin-like (ANGPTL) protein family. Encoded by the ANGPTL6 gene in humans, this protein was first identified and characterized based on its structural homology to other angiopoietin family members, yet it shows unique biological functions that are largely independent of angiogenesis. Unlike classic angiopoietins that mainly regulate blood vessel formation and stability, ANGPTL6 has emerged as a critical endocrine factor involved in systemic metabolism, energy homeostasis, inflammation, and cellular stress responses. It is synthesized and secreted into the bloodstream, allowing it to act on multiple distant tissues and organs to coordinate physiological and pathological processes. Due to its broad functional spectrum and strong association with metabolic diseases, ANGPTL6 has become an important molecule in biomedical research related to obesity, diabetes, cardiovascular disorders, and age-related diseases.

Regulatory Role of ANGPTL6 in Lipid Metabolism
One of the best-characterized functions of ANGPTL6 is its potent regulatory effect on lipid metabolism, especially through modulating lipoprotein lipase (LPL) activity. LPL is a rate-limiting enzyme that catalyzes the hydrolysis of triglycerides in circulating lipoproteins, releasing free fatty acids for tissue uptake or storage. ANGPTL6 acts as an endogenous inhibitor of LPL, reducing its enzymatic activity and slowing down triglyceride breakdown. By inhibiting LPL in different tissues, ANGPTL6 regulates plasma triglyceride levels, lipid distribution, and fatty acid availability for energy production. Dysregulation of ANGPTL6 expression is closely linked to hypertriglyceridemia, a major risk factor for atherosclerosis and cardiovascular diseases. Moreover, ANGPTL6 also affects cholesterol metabolism and fatty acid oxidation in the liver and muscle, making it a central regulator of whole-body lipid homeostasis and energy balance.
Potential Clinical Applications and Therapeutic Perspectives of ANGPTL6
Due to its critical roles in metabolism and disease, ANGPTL6 has attracted growing attention for its potential clinical applications, including biomarker use and therapeutic development. As a circulating factor, ANGPTL6 can be easily detected in blood samples, making it a promising non-invasive biomarker for early screening of metabolic syndrome, type 2 diabetes, and cardiovascular risk. For therapeutic development, strategies targeting ANGPTL6, such as neutralizing antibodies, antisense oligonucleotides, or small-molecule inhibitors, are being explored to reduce its expression or block its activity. These approaches aim to restore LPL function, improve lipid profiles, enhance insulin sensitivity, and reduce cardiovascular risk. Further research is needed to clarify the precise signaling pathways, receptors, and tissue-specific effects of ANGPTL6, which will facilitate the translation of basic research findings into clinical tools for disease prevention and treatment.
