"Abstract: Viticulture and associated products are an important part of the economy in many countries. However, biotic and abiotic stresses impact negatively the production of grapes and wine. Climate change is in many aspects increasing both these stresses. Routine sample retrievals and analysis tend to be time-consuming and require expensive equipment and skilled personnel to operate. These challenges could be overcome through the development of a miniaturized analytic device for early detection of grapevine stresses in the field. Abscisic acid is involved in several plant processes, including the onset of fruit ripening and tolerance mechanisms against drought stress. This hormone can be detected through a competitive immunoassay and is found in plants in concentrations up to 10-1 mg/mL. A microfluidic platform is developed in this work which can detect a minimum of 10-11 mg/mL of abscisic acid in buffer. Grape samples were tested using the microfluidic system alongside benchmark techniques such as high-performance liquid chromatography. The microfluidic system could detect the increase to 10-5 mg/mL of abscisic acid present in real berry samples at the veraison stage of ripening."

"Abstract:Point-of-care (PoC) devices offer a promising solution for fast, portable, and easy-to-use diagnostics. These characteristics are particularly relevant in agrifood fields like viticulture where the early detection of plant stresses is crucial to crop yield. Microfluidics, with its low reagent volume requirements, is well-suited for such applications. Self-driven microfluidic devices, which rely on capillary forces for fluid motion, offer an attractive alternative by eliminating the need for external pumps and complex fluid control systems. However, traditional microfluidic prototyping materials like polydimethylsiloxane (PDMS) present challenges due to their hydrophobic nature. This paper presents the development of a reusable, portable, capillary-driven microfluidic platform based on a PDMS-PEG (polyethylene glycol) copolymer designed for the rapid low-cost detection of abscisic acid (ABA), a key biomarker for the onset of ripening of non-climacteric fruits and drought stress in vines. By employing passive fluid transport mechanisms, such as capillary-driven sequential flow, this platform enables precise biological and chemical screenings while maintaining portability and ease of use. A simplified field-ready sample processing method is used to prepare the grapes for analysis."

The relationship between fluorescence signal intensity and ABA concentration in treated table grapes was evaluated, with the results presented in Figure 7. Origin of Weakened Interannual Sea Surface Temperature Variability in the Southeastern Tropical Atlantic Ocean

Abstract

Observations and reanalysis products are used to investigate the substantial weakening in the southeastern tropical Atlantic sea surface temperature (SST) variability since 2000. Relative to 1982-1999, the March-April-May SST variability in the Angola-Benguela area (ABA) has decreased by more than 30%. Both equatorial remote forcing and local forcing are known to play an important role in driving SST variability in the ABA. Compared to 1982-1999, since 2000, equatorial remote forcing had less influence on ABA SSTs, whereas local forcing has become more important. In particular, the robust correlation that existed between the equatorial zonal wind stress and the ABA SSTs has substantially weakened, suggesting less influence of Kelvin waves on ABA SSTs. Moreover, the strong correlation linking the South Atlantic Anticyclone and the ABA SSTs has reduced. Finally, multidecadal surface warming of the ABA could also have played a role in the weakening of the interannual SST variability.

Genome-wide identification and expression patterns analysis of theRPD3/HDA1gene family in cotton

Abstract

Background Histone deacetylases (HDACs) catalyze histone deacetylation and suppress gene transcription during various cellular processes. Within the superfamily of HDACs,RPD3/HDA1-type HDACs are the most studied, and it is reported thatRPD3genes play crucial roles in plant growth and physiological processes. However, there is a lack of systematic research on theRPD3/HDA1gene family in cotton. Results In this study, genome-wide analysis identified 9, 9, 18, and 18RPD3genes inGossypium raimondii, G. arboreum,G. hirsutum, andG. barbadense, respectively. This gene family was divided into 4 subfamilies through phylogenetic analysis. The exon-intron structure and conserved motif analysis revealed high conservation in each branch of the cottonRPD3genes. Collinearity analysis indicated that segmental duplication was the primary driving force during the expansion of theRPD3gene family in cotton. There was at least one presumed cis-element related to plant hormones in the promoter regions of allGhRPD3genes, especially MeJA- and ABA-responsive elements, which have more members than other hormone-relevant elements. The expression patterns showed that mostGhRPD3genes had relatively high expression levels in floral organs and performed higher expression in early-maturity cotton compared with late-maturity cotton during flower bud differentiation. In addition, the expression ofGhRPD3genes could be significantly induced by one or more abiotic stresses as well as exogenous application of MeJA or ABA. Conclusions Our findings reveal thatGhRPD3genes may be involved in flower bud differentiation and resistance to abiotic stresses, which provides a basis for further functional verification ofGhRPD3genes in cotton development and a foundation for breeding better early-maturity cotton cultivars in the future.
