NALFIA is a general device that combines lateral flow technology and immunoassay principle for detection of nucleic acid related products.
Creative Diagnostics provides a series of antibodies against nucleic acid labels that are used extensively in NALFIA. These antibodies can specifically bind to small molecule labels and facilitate the development of NALF methods.
Conventional nucleic acid detection methodologies require well-trained operators and sophisticated apparatus for operation, with signal readout and detection times often taking several hours. With increasing demand for diagnostics under resource-limited conditions, lateral flow assays (LFAs) for nucleic acid testing have gained greater attention for their low-costs and easy-to-use operations. Likewise, they have shown great potential in the point-of-care testing (POCT) sector.
Amplification of target genes has been widely used in diagnostic frameworks. Among them, polymerase chain reaction (PCR) and the isothermal methods including loopmediated isothermal amplification (LAMP), multiple cross displacement amplification (MCDA), and recombinase polymerase amplification (RPA) are introduced on LFIA, called nucleic acid lateral flow immunoassay (NALFIA).

A typical workflow of lateral flow nucleic testing comprises nucleic acid extraction from biological samples, nucleic acid amplification, and the detection of amplicons through LFAs using colorimetric labeling materials.
For NALFIA, both forward and reverse primers are typically tagged with biotin and another antigen with high antibody affinity, i.e. digoxigenin (DIG), carboxyfluorescein (FAM) and/or FITC. Thus, amplicons are dual-hapten labeled, and form sandwich-type complexes via high affinity binding between antigen and antibody immunoreactions. Each monoclonal antibody can specifically recognize the corresponding tag.
