POLDIP2 Full Name
polymerase (DNA-directed), delta interacting protein 2
POLDIP2 Introduction
Polymerase delta-interacting protein 2 (POLDIP2), also known as PDIP38, is an exceptionally versatile, multi-compartmental protein initially discovered through its direct interaction with DNA polymerase delta and PCNA in the cell nucleus. While it contributes to DNA replication and translesion synthesis, POLDIP2 is most renowned for its profound physiological functions outside the nucleus. In the cytoplasm, it localizes to mitochondria to regulate energy dynamics and strongly associates with the actin cytoskeleton and focal adhesions. Crucially, POLDIP2 is a primary positive regulator of NADPH oxidase 4 (NOX4). By directly binding to and stabilizing NOX4, POLDIP2 actively stimulates the controlled production of reactive oxygen species (ROS), specifically hydrogen peroxide. This localized ROS production acts as a vital secondary messenger pathway to heavily influence cytoskeletal remodeling, cellular migration, and vascular tone.

The clinical significance of POLDIP2 spans across severe cardiovascular diseases and oncology. In the vascular system, abnormal upregulation of POLDIP2 drives excessive ROS production via NOX4, leading to severe vascular smooth muscle hypertrophy, arterial stiffness, and the pathogenesis of life-threatening aortic aneurysms. Conversely, in the context of oncology, POLDIP2 acts as a potent driver of tumor metastasis. Highly aggressive solid tumors, such as breast cancer and glioblastoma, frequently overexpress POLDIP2. This overexpression hyperactivates focal adhesion kinase (FAK) and RhoA pathways, drastically enhancing the physical contractility and invasive capacity of cancer cells. By physically equipping cancer cells to dismantle extracellular barriers, POLDIP2 facilitates rapid dissemination and metastasis, marking it as a compelling therapeutic target for both vascular aneurysms and metastatic carcinomas.
