PLA2G1B
PLA2G1B Full Name
phospholipase A2, group IB (pancreas)
PLA2G1B Introduction
Phospholipase A2, group IB (PLA2G1B), conventionally known as pancreatic phospholipase A2, is a highly specific, secreted digestive enzyme synthesized predominantly by the pancreatic acinar cells. Following its secretion into the small intestine in an inactive zymogen form, it is rapidly activated by trypsin. Physiologically, the classical and primary function of PLA2G1B is to aggressively hydrolyze dietary phospholipids—specifically targeting the sn-2 ester bond—to produce essential free fatty acids and lysophospholipids. This biochemical degradation is an absolute prerequisite for the efficient intestinal absorption of complex dietary fats and fat-soluble vitamins, making PLA2G1B a cornerstone of mammalian lipid metabolism and systemic nutritional homeostasis.

Beyond its canonical role in benign intestinal digestion, the systemic impact of PLA2G1B is profoundly implicated in the modern epidemic of metabolic syndrome. Extensive translational research has revealed that PLA2G1B is a critical driver of diet-induced obesity, insulin resistance, and severe hepatosteatosis (fatty liver). When individuals consume a modern, high-fat Western diet, the hyper-efficient lipid absorption mediated by PLA2G1B floods the systemic circulation with excess lysophospholipids. These specific lipid metabolites act as potent systemic signaling molecules that actively suppress tissue energy expenditure and heavily promote adipocyte hypertrophy (fat cell enlargement). Consequently, the targeted pharmacological inhibition of PLA2G1B in the gastrointestinal tract has emerged as a highly promising, non-systemic therapeutic intervention. By deliberately inducing controlled phospholipid malabsorption, PLA2G1B inhibitors can effectively shield individuals from diet-induced severe obesity, completely reverse insulin resistance, and halt the progression of metabolic diseases without affecting the central nervous system.
