Nid1 Introduction
Nidogen-1(NID-1),formerly known as entactin, it is a protein encoded by the NID1 gene in humans. Nestinogen-1 and Nestinogen-2 are important components of the basement membrane, alongside other components such as type IV collagen, proteoglycans (heparan sulfate and glycosaminoglycans), laminin, and fibronectin. Nidogen 1 (historically also known as Entactin) is a sulfated glycoprotein of~150 kDa, which is a core structural component commonly found in all basement membranes (BMs). BM is a special sheet-like extracellular matrix located beneath the epithelial and endothelial cell layers, surrounding muscle fibers, adipocytes, and nerves. Functionally speaking, Nidogen 1 is best described as a key "connector" or "bridge" molecule. Its main and decisive role is to connect the two main, independently assembled scaffold networks of the basement membrane: the laminin network and the type IV collagen network. This bridging activity is the foundation for the integrity, stability, and proper assembly of BM structures.

Function
Nidogen 1 is a member of the Nidogen family of basement membrane glycoproteins. This protein interacts with several other components of the basement membrane. Structurally, it connects the network formed by collagen and laminin together with perlite. It may also play a role in the interaction between cells and extracellular matrix. Insulin precursor 1 is the main organizer and stabilizer of the basement membrane. Although its bridging function is partially redundant with Nidogen 2, it is crucial for creating a complete and functional BM that is essential for tissue development and homeostasis. Tumors and stromal cells secrete proteases that specifically cleave Nidogen 1, such as MMP-2, MMP-3, MMP-7, MMP-9. This degradation weakens the BM barrier and promotes local invasion and metastasis. The Nidogen 1 fragment in body fluids has been studied as a potential cancer biomarker.
