MEF2D
MEF2D Full Name
myocyte enhancer factor 2D
MEF2D Introduction
MEF2D (myocyte enhancer factor 2D) is a MADS-box transcription factor of the MEF2 family that, unlike its relatives, is broadly expressed across both muscle and non-muscle lineages. The protein binds A/T-rich DNA elements as a homodimer or as a heterodimer with other MEF2 isoforms, and it integrates calcium- and MAPK-dependent signaling inputs through its transactivation domain. The MEF2D gene lies on chromosome 1q22 and gives rise to several isoforms whose relative abundance shifts during neuronal maturation and cardiac development.

Transcriptional Regulation in Muscle, Neurons, and Immune Cells
MEF2D partners with the basic helix–loop–helix transcription factor MyoD to activate skeletal muscle differentiation genes, and it cooperates with GATA4 and TBX5 to drive cardiac gene expression in cardiomyocytes. In neurons, MEF2D regulates activity-dependent genes such as Arc and SynGAP, and its sumoylation and disinhibition from Cabin1–HDAC complexes are required for memory consolidation. MEF2D also has important roles outside of muscle and brain: in B cells it cooperates with MEF2B to drive germinal-center programs, and in chondrocytes it promotes hypertrophic differentiation downstream of BMP signaling.
Neurodegeneration, Autism, and Leukemia
MEF2D has been identified as a key pro-survival factor for cerebellar granule neurons, and its overexpression rescues neurodegeneration in models of spinocerebellar ataxia type 1. Rare de novo mutations in MEF2D are reported in patients with autism spectrum disorder and intellectual disability, where dysregulated activity-dependent transcription alters excitatory synapse development. A recurrent MEF2D–DAZAP1 fusion gene generated by chromosome 1 translocations is a hallmark of B-cell precursor acute lymphoblastic leukemia, and the fusion protein drives leukemogenesis through aberrant MEF2-dependent transcriptional programs.
