IL3 Introduction
Interleukin-3 (IL-3) is a pleiotropic hematopoietic cytokine primarily produced by activated T cells and mast cells, playing a central role in early blood cell development and immune system regulation. It acts on hematopoietic stem and progenitor cells, particularly CD34+ populations, to support proliferation and differentiation across multiple myeloid lineages. However, emerging evidence highlights a paradox in its biological activity: while IL-3 can expand progenitor populations under short-term culture conditions, it may simultaneously compromise long-term stem cell self-renewal capacity, especially in transplantation or ex vivo expansion settings. This creates a critical challenge for researchers aiming to optimize stem cell therapies, where balancing expansion efficiency with functional stemness remains a major bottleneck.

In disease contexts, dysregulated IL-3 signaling is increasingly linked to both inflammatory and malignant conditions. Elevated IL-3 expression has been associated with chronic inflammatory disorders such as inflammatory bowel disease (IBD), where it influences regulatory T cell (Treg) biomechanics and migration, thereby shaping immune responses in inflamed tissues. In hematological malignancies, IL-3 signaling contributes to aberrant survival and proliferation pathways, particularly through its receptor CD123. This axis is frequently implicated in acute myeloid leukemia (AML), blastic plasmacytoid dendritic cell neoplasm (BPDCN), and other myeloproliferative disorders. Clinically, IL-3-driven signaling networks can reinforce tumor cell persistence and immune evasion, making it a key contributor to disease progression and treatment resistance in blood-related cancers.
From a translational perspective, IL-3 signaling has become an important therapeutic and bioengineering target due to its involvement in JAK2/STAT5 and MAPK pathway activation following receptor engagement. The IL-3 receptor system, particularly CD123, is now widely explored for targeted therapies including CAR-T cell approaches and IL-3–based immunotoxins, reflecting its high relevance in precision oncology. At the same time, recent discoveries showing IL-3 interactions with viral capsids and engineered delivery systems suggest an additional role in modulating biologic transport and immune recognition, potentially impacting gene therapy vector design. Together, these findings position IL-3 not only as a key immunoregulatory cytokine but also as a critical node in emerging therapeutic strategies aimed at both hematologic disease control and advanced drug delivery optimization.
