FSCN1 Introduction
Fascin actin-bundling protein 1 (FSCN1), encoded by the FSCN1 gene, is a highly conserved 55-kDa cytoskeletal protein that functions as the primary organizer of actin filaments into tight, parallel bundles. Structurally, the protein adopts a unique β-trefoil fold, which allows it to fit snugly within the grooves of actin filaments. Unlike other actin cross-linkers (such as α-actinin) that form loose networks, FSCN1 packs F-actin into rigid, stiff bundles. These bundles are the structural core of filopodia (finger-like projections) and invadopodia (invasive structures). Physiologically, these protrusions are the "legs" and "sensors" of the cell; they are essential for cell migration, neurite outgrowth during development, and the formation of cell-matrix adhesions, enabling cells to explore and move through their environment.

The clinical importance of FSCN1 is almost exclusively tied to its role as a potent promoter of cancer metastasis. While FSCN1 expression is low or absent in most normal adult epithelial tissues, it is drastically upregulated in high-grade carcinomas, including breast, colon, lung, and esophageal cancers. This upregulation is often a downstream effect of the Epithelial-to-Mesenchymal Transition (EMT) program. By equipping tumor cells with abundant filopodia and invadopodia, FSCN1 enhances their motility and invasiveness, allowing them to breach basement membranes and disseminate to distant organs. Consequently, high levels of FSCN1 are clinically correlated with aggressive tumor phenotypes, increased risk of metastasis, and significantly shorter patient survival rates (poor prognosis), identifying it as a promising therapeutic target for blocking metastasis.
