FBXO17 Full Name
F-box protein 17
FBXO17 Introduction
FBXO17 (F-box protein 17) is a member of the F-box protein family that functions as a substrate-recognition component of the SCF (SKP1-CUL1-F-box) E3 ubiquitin ligase complex. For researchers and clinicians trying to understand why cancer cells sustain abnormal signaling despite regulatory checkpoints, FBXO17 is increasingly discussed as part of the ubiquitin–proteasome system that governs selective protein degradation and maintains intracellular protein homeostasis. By determining which proteins are tagged for degradation, F-box proteins such as FBXO17 indirectly shape key cellular outcomes including survival, stress adaptation, and fate decisions. Within the broader landscape of cancer biology, F-box proteins are now recognized as multilayered regulators acting not only within tumor cells but also across the tumor microenvironment, placing FBXO17 within a growing category of proteostasis-related targets that may help explain persistent therapeutic resistance and disease progression.

Functionally, FBXO17 has been implicated in several oncogenic processes that directly address major clinical challenges such as uncontrolled proliferation, metastasis, and metabolic adaptation. In glioma models, FBXO17 is frequently upregulated and has been associated with enhanced tumor cell growth, migration, invasion, and epithelial–mesenchymal transition (EMT), a key biological program driving metastatic dissemination and treatment resistance. Mechanistically, FBXO17 appears to promote these malignant behaviors through activation of the Akt/GSK-3β/Snail signaling axis, a pathway widely known for its role in EMT regulation and tumor aggressiveness. In addition to signaling control, FBXO17 also contributes to metabolic reprogramming by enhancing glycolytic activity, supporting increased glucose uptake, ATP production, and lactate accumulation, along with elevated expression of key glycolysis-related enzymes. This dual influence on both signaling and metabolism positions FBXO17 as a functional coordinator of tumor adaptation under conditions of rapid growth and environmental stress.
From a disease perspective, FBXO17 is most strongly associated with high-grade glioma, where elevated expression correlates with more aggressive clinical behavior and poorer outcomes. Experimental suppression of FBXO17 has been shown to reduce tumor cell proliferation, invasion, and migration, while increasing apoptosis and oxidative stress, suggesting a dependency of glioma cells on this regulatory axis for survival and progression. These findings highlight FBXO17 as a potential vulnerability in tumors characterized by strong metabolic demands and EMT-driven invasiveness. More broadly, emerging research on F-box protein networks and microRNA-mediated regulation of the ubiquitin–proteasome system suggests that FBXO17 may also participate in upstream regulatory circuits that fine-tune oncogenic signaling. Although its full clinical potential is still being defined, FBXO17 is increasingly viewed as a promising mechanistic link between proteostasis imbalance, metabolic rewiring, and tumor progression, making it a relevant focus for future cancer biomarker and therapeutic strategy development.
