ELAVL4
ELAVL4 Full Name
ELAV (embryonic lethal, abnormal vision, Drosophila)-like 4 (Hu antigen D)
ELAVL4 Introduction
ELAVL4 (also known as HuD) is a neuron-specific RNA-binding protein belonging to the ELAV/Hu family, distinguished from its ubiquitously expressed paralogs (HuR/HuB/HuC) by its near-exclusive expression in the central and peripheral nervous system. The protein contains three RNA recognition motifs (RRMs); the N-terminal tandem RRMs bind AU-rich elements with high affinity while the third RRM contributes to cooperative binding on long 3′UTRs. In humans, ELAVL4 maps to chromosome 1p33–p32.3, a region notable for its association with Parkinson disease susceptibility, and its transcripts undergo extensive alternative splicing that gives rise to isoforms with subtly different mRNA-stabilizing capacities in differentiating neurons.

Neuronal mRNA Stability and Transport
HuD binds AU-rich elements in the 3′UTRs of mRNAs encoding GAP-43, NeuN, and several neuronal transcription factors, prolonging their half-lives in developing axons and dendrites. Through its RRM domains, ELAVL4 also assembles into ribonucleoprotein granules that shuttle specific transcripts from the soma to distal neurites, a process that supports local translation during axon outgrowth and synaptic remodeling. Loss of ELAVL4 in murine models reduces neurite complexity, while its overexpression enhances regenerative responses after nerve injury. Recent work further implicates ELAVL4 in regulating alternative splicing and polyadenylation decisions through interactions with SRSF and CELF proteins.
Neurological Disease and Paraneoplastic Syndromes
ELAVL4 was originally identified as the antigen targeted by anti-Hu antibodies in patients with paraneoplastic encephalomyelitis, an autoimmune complication of small-cell lung cancer in which neurons are attacked because tumor cells ectopically express HuD. Genetic variants near the ELAVL4 locus have also been linked to late-onset Parkinson disease in multiple GWAS, although the functional consequence of these variants is still debated. In spinal muscular atrophy and amyotrophic lateral sclerosis, dysregulated HuD levels affect SMN and neurofilament mRNA processing, raising the possibility that modulating ELAVL4 expression could stabilize motor neuron transcripts.
