CCNE1
CCNE1 Full Name
cyclin E1
CCNE1 Introduction
CCNE1 is a key gene located on human chromosome 19q12, encoding the Cyclin E1 protein. It belongs to the highly conserved family of cell cycle proteins, whose members are key regulators of the cell cycle, which is an ordered process of cell growth and division. Cyclin E has been identified as a G1/S phase specific cyclin in mammalian cells. When researchers discovered that its gene CCNE1 was overexpressed, amplified, or dysregulated in various human cancers, it quickly attracted widespread attention. This establishes that CCNE1 is not only a core cell cycle component, but also a powerful oncogene. When dysregulated, it drives uncontrolled cell proliferation, which is a hallmark of cancer and makes it a focus of oncology research.

CCNE1 is a fundamental regulator of the cell cycle, and its normal periodic function is crucial for healthy cell division. It frequently undergoes dysregulation through amplification or overexpression in various cancers, transforming it into a powerful oncogenic driver, promoting uncontrolled proliferation and genomic disorder. This established role makes CCNE1 not only a valuable prognostic biomarker, but also a promising therapeutic target. The ongoing research on CDK2 inhibitors and other novel drugs has great potential to improve the prognosis of cancer patients driven by CCNE1.
Targeted Therapy
The core role of dysregulated CCNE1-CDK2 activity in driving cancer makes it a compelling therapeutic target. The current clinical development strategies include:
CDK2 inhibitor: a selective small molecule inhibitor designed to block the kinase activity of Cyclin E1-CDK2 complex.
CCNE1 amplification of CDK4/6 inhibitors in cancer: This study explores the interaction between the cyclin D-CDK4/6 and cyclin E-CDK2 pathways and potential combination therapy.
PROTACs (Protein Hydrolysis Targeted Chimeras): A novel molecule designed to specifically label the cyclin E1 protein for degradation by the cell's own proteasome system.
Alternate Names for CCNE1
CCNE1; cyclin E1; CCNE; G1/S-specific cyclin-E1; cyclin Es; cyclin Et; cyclin E variant ex5del; cyclin E variant ex7del;
