BLK Full Name
B lymphoid tyrosine kinase
BLK Introduction
BLK (B lymphoid tyrosine kinase) is a non-receptor protein tyrosine kinase belonging to the Src family of kinases (SFKs). The gene encoding BLK is located on human chromosome 8p23.1 and produces a protein of approximately 505 amino acids with a molecular weight of about 55-60 kDa. Unlike many other Src family kinases that are ubiquitously expressed, BLK exhibits a highly restricted expression pattern, primarily found in B lymphocytes, as well as in some other immune cell types and in pancreatic beta cells. BLK plays critical roles in B cell receptor (BCR) signaling, B cell development, and B cell activation. The kinase is involved in the early stages of BCR signal transduction, where it phosphorylates immunoreceptor tyrosine-based activation motifs (ITAMs) on the BCR-associated signaling subunits Igα (CD79a) and Igβ (CD79b). BLK also contributes to the regulation of insulin secretion in pancreatic beta cells, linking it to metabolic disorders such as type 2 diabetes. Genetic variants in the BLK gene have been associated with autoimmune diseases (including systemic lupus erythematosus and rheumatoid arthritis) and metabolic traits.

Enzymatic Function and Signaling Mechanisms
BLK functions as a protein tyrosine kinase that phosphorylates specific tyrosine residues on its target proteins. The kinase activity is tightly regulated by the phosphorylation state of the C-terminal regulatory tyrosine (Tyr497) and by interactions with SH2 and SH3 domain ligands. In B cells, BLK is one of several Src family kinases (including LYN, FYN, and LCK) that initiate BCR signaling. Upon BCR engagement by antigen, BLK (along with LYN) phosphorylates the ITAMs within the cytoplasmic tails of Igα and Igβ. The phosphorylated ITAMs then recruit and activate the tyrosine kinase SYK (spleen tyrosine kinase) via its SH2 domains.
Role in B Cell Development and Immune Function
BLK is expressed at specific stages of B cell development, primarily in immature and mature B cells, but is downregulated in plasma cells. Studies using Blk knockout mice have provided insights into its physiological functions. Blk⁻/⁻ mice are viable and fertile but exhibit subtle defects in B cell development and function. They have reduced numbers of mature B cells in the peripheral lymphoid organs, impaired BCR-induced proliferation, and decreased antibody responses to T-cell-dependent antigens. However, the phenotype is milder than that of mice lacking other Src family kinases (such as Lyn), suggesting partial redundancy among these kinases.
