ARHGEF1 Full Name
Rho guanine nucleotide exchange factor (GEF) 1
ARHGEF1 Introduction
ARHGEF1 (Rho guanine nucleotide exchange factor 1, also known as p115-RhoGEF or LBC) is a member of the Dbl family of RhoGEFs that activate Rho family small GTPases by catalyzing the exchange of GDP for GTP. The gene is located on chromosome 19q13.2 and encodes a protein of 912 amino acids with a molecular weight of approximately 115 kDa. ARHGEF1 is ubiquitously expressed in human tissues, with particularly high levels in the brain, lung, kidney, and hematopoietic cells. The protein functions as a key regulator of the RhoA signaling pathway, specifically activating RhoA and, to a lesser extent, RhoB and RhoC. ARHGEF1 is unique among RhoGEFs as it contains a regulator of G protein signaling (RGS) domain that directly interacts with and is activated by the Gα12 and Gα13 subunits of heterotrimeric G proteins, linking G protein-coupled receptor (GPCR) signaling to RhoA activation. Through this mechanism, ARHGEF1 mediates cellular responses to various extracellular signals including lysophosphatidic acid (LPA), thrombin, angiotensin II, and sphingosine-1-phosphate (S1P). ARHGEF1 regulates diverse cellular processes including actin cytoskeleton reorganization, stress fiber formation, focal adhesion assembly, cell contraction, cell migration, and gene expression. Dysregulation of ARHGEF1 has been implicated in various human diseases including cancer, cardiovascular disease, and neurological disorders.

Pathophysiological and Clinical Significance of ARHGEF1
Sustained overactivation, aberrant expression or functional deficiency of ARHGEF1 is a key driving factor for the development of inflammatory diseases, vascular injuries, neurological disorders and malignancies, with important diagnostic and therapeutic implications. In chronic inflammatory diseases, excessive ARHGEF1 activity continuously activates Rho signaling, amplifying immune cell infiltration and pro-inflammatory cytokine release, which aggravates tissue damage in rheumatoid arthritis, inflammatory bowel disease and systemic inflammatory response syndrome. In vascular diseases, abnormal ARHGEF1 activation disrupts endothelial barrier integrity, increasing vascular permeability and promoting the progression of hypertension, atherosclerosis and acute vascular injury. In human cancers, ARHGEF1 is frequently upregulated in lung cancer, breast cancer and gastrointestinal tumors; it enhances tumor cell motility and invasiveness by overactivating Rho cascades, facilitating tumor metastasis and angiogenesis. Clinically, ARHGEF1 expression level is closely correlated with disease severity and tumor prognosis, serving as a valuable prognostic marker and a potential target for targeted therapy.
Expression Regulation and Transcriptional Control of ARHGEF1
The transcription, protein abundance and activity of ARHGEF1 are precisely regulated by transcription factors, post-transcriptional silencing and epigenetic modifications. Ubiquitous and tissue-specific transcription factors bind to the promoter region to maintain stable basal expression in normal tissues. When cells receive inflammatory, proliferative or stress stimuli, transcriptional upregulation rapidly elevates ARHGEF1 expression to meet enhanced signaling demands. Multiple microRNAs target the 3'-UTR of ARHGEF1 mRNA to inhibit translation and accelerate mRNA degradation, acting as a post-transcriptional brake to prevent excessive protein accumulation. Epigenetic modifications including DNA methylation and histone acetylation control long-term gene expression patterns. Abnormal epigenetic modification in diseased tissues leads to persistent ARHGEF1 overexpression, which sustains pathological signaling and accelerates disease progression.
