ACTB Introduction
β-actin is encoded by the human ACTB gene and is one of the six highly conserved subtypes of actin found in mammals. It is the main component of the cytoskeleton and is primarily expressed in non muscle cells. As a core participant in cellular structure and movement, β - actin is often regarded as a "housekeeping" protein because it is necessary and consistently expressed in almost all eukaryotic cell types. Its richness and stability also make it a typical load control in molecular biology experiments. Actin β has been shown to interact with SPTBN2. [8] In addition, it was found that RNA binding protein Sam68 interacts with mRNA encoding actin β, which regulates synaptic formation of dendritic spines through its cytoskeletal components. Actin is a highly conserved protein involved in cell movement, structure, and integrity. Alpha actin is the main component of the contractile apparatus.

Application
Actin beta is commonly used in Western blotting as a load control to normalize total protein mass and examine the final protein degradation in the sample. Its transcript is also commonly used as a housekeeping gene standard in qPCR. Its molecular weight is approximately 42 kDa. Due to its constitutive and high-level expression, β - actin is widely used as an internal control in techniques such as Western blotting, qRT PCR, and immunofluorescence to normalize changes in protein or RNA loading, transfer efficiency, and cell count. However, under certain experimental conditions (such as drug therapy, differentiation), its expression may vary, which requires validation and sometimes the use of alternative controls. The newly discovered heterozygous missense mutation in the ACTB gene leads to Baraitser Winter's frontoparietal syndrome, characterized by distinct facial features, brain deformities, and intellectual disability. These mutations disrupt actin polymerization and the integrity of the cytoskeleton.
Clinical Relevance
Recurrent mutations in this gene have been associated to cases of diffuse large B-cell lymphoma.De novo gain-of-function mutations in this gene is associated with Baraitser-Winter syndrome.Beta-actin is far more than a static scaffold; it is a dynamic, information-processing platform essential for life.
