ABCA6 Introduction
ATP-binding cassette sub-family A member 6 (ABCA6) is a full-size transmembrane protein belonging to the highly conserved ATP-binding cassette (ABC) transporter superfamily. It is a defining member of the "ABCA6-like" transporter evolutionary subgroup—which also includes ABCA8, ABCA9, and ABCA10—clustered together on human chromosome 17q24. Structurally, ABCA6 is composed of two tandemly oriented halves, each containing a transmembrane domain (TMD) for substrate translocation and a highly conserved intracellular nucleotide-binding domain (NBD) that powers the transport process through ATP hydrolysis. ABCA6 is ubiquitously expressed but exhibits particularly high mRNA levels in the liver, lung, heart, brain, and monocyte-derived macrophages.

ABCA6 plays a significant role in intracellular lipid transport and the maintenance of cellular cholesterol homeostasis. In human macrophages, its expression is tightly regulated by lipid metabolism; it is markedly upregulated during the differentiation of monocytes into macrophages, yet its mRNA levels are dynamically responsive to cellular cholesterol loading. Beyond macrophages, in vascular endothelial cells, the transcription of ABCA6 is directly regulated by Forkhead box O (FoxO) transcription factors. By utilizing the energy derived from ATP, ABCA6 is believed to facilitate the intracellular trafficking and dynamic redistribution of specific lipid molecules or cholesterol derivatives across internal organelle membranes, ensuring proper cellular lipid balance.
Clinically, because of its intimate involvement in macrophage lipid handling, the functional dysregulation of ABCA6 is closely linked to metabolic and cardiovascular pathologies. Disruptions in macrophage cholesterol transport can lead to the excessive intracellular accumulation of esterified cholesterol, promoting the formation of lipid-laden "foam cells"—a critical and defining early step in the pathogenesis of atherosclerosis and coronary artery disease. Furthermore, aberrant expression patterns of ABCA6 have been documented in certain oncological contexts, such as mantle cell lymphoma, suggesting that alterations in intracellular lipid transport driven by ABCA6 may contribute to tumor cell survival and disease pathogenesis.
