CDP-diacylglycerol--glycerol-3-phosphate 3-phosphatidyltransferase; Phosphatidylglycerophosphate synthase (PGP synthase); pgsA; MW1166
Purity
> 85% as determined by SDS-PAGE
Format
Liquid or Lyophilized powder
Concentration
Batch dependent - please inquire should you have specific requirements.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Storage
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL. We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Introduction
Staphylococcus aureus CDP-diacylglycerol--glycerol-3-phosphate 3-phosphatidyltransferase (TarD) is a key enzyme in cell wall biosynthesis, catalyzing the formation of CDP-glycerol from glycerol-3-phosphate and CTP. This reaction initiates the assembly of teichoic acid polymers, essential components covalently linked to peptidoglycan that maintain cell envelope integrity in Gram-positive bacteria. TarD exhibits unique structural properties—despite high sequence similarity to homologs like Bacillus subtilis TagD, it forms tetramers (in addition to dimers) and displays distinct kinetic mechanisms. Its disordered C-terminal region, contrasting with TagD's helical structure, underpins these functional differences. As teichoic acids are vital for bacterial viability, TarD represents a promising antibiotic target, especially against drug-resistant S. aureus strains.
