General Description
Bispecific antibodies (BsAbs) are engineered immunoglobulins that simultaneously bind two distinct antigens or two different epitopes on the same antigen. Their function depends on precise heavy- and light-chain pairing and molecular architecture, which determine avidity, orientation, and functional potency. By physically bridging immune effector cells to target cells, BsAbs can redirect and amplify immune-mediated cytotoxicity, offering enhanced tumor killing and a reduced likelihood of resistance compared with some monospecific therapies B-lymphocyte antigen CD19 is also known as CD19 (Cluster of Differentiation 19), is a single-pass type I membrane protein which contains two Ig-like C2-type (immunoglobulin-like) domains. CD19 is expressed on follicular dendritic cells and B cells. Mutations in CD19 are associated with severe immunodeficiency syndromes characterized by diminished antibody production. CD19 has been shown to interact with: CD81, CD82, Complement receptor 2, and VAV2. CD3 (cluster of differentiation 3) is a protein complex and T cell co-receptor that is involved in activating both the cytotoxic T cell (CD8+ naive T cells) and T helper cells (CD4+ naive T cells). It is composed of four distinct chains In mammals, the complex contains a CD3γ chain, a CD3δ chain, and two CD3s chains. These chains associate with the T-cell receptor (TCR) and the CD3-zeta (ζ-chain) to generate an activation signal in T lymphocytes. The TCR. CD3-zeta, and the other CD3 molecules together constitute the TCR complex. A CD19 & CD3 bridging ELISA evaluates a bispecific molecule's ability to form the ternary complex between CD19 and CD3, providing a direct measure of the antibody's primary mechanism ofaction
