Introduction
Hcp1 is a core component and substrate of the Type VI Secretion System cluster-1 (T6SS-1) in Burkholderia mallei, which is essential for virulence, intracellular survival, and cell-cell spread. It forms part of the secretion tube of T6SS-1, together with VgrG, and is secreted into culture supernatants and during infection. Hcp1 contributes to the formation of multinucleated giant cells (MNGCs) in infected macrophages and supports actin-based motility and intercellular spread. Its expression is regulated by environmental cues and regulatory systems such as VirAG, and it is highly induced after bacterial uptake in host cells. Because of its immunogenicity and critical role in pathogenesis, Hcp1 is being considered as a vaccine target or diagnostic marker.
