Contents of Kit
Warning: Do not use any reagents where damage to the packaging has occurred.
The ELISA contains sufficient reagents for 96 wells. Each kit contains the following reagents:
1. Coated Microtiter Strips: ELISA plate strip holder with 96 (12×8 strips) coated polystyrene microtiter wells. Store at 2-8°C until expiry.
2. Sample Dilution Buffer: One bottle, 25 mL, to be used for preparing sample dilutions. A slight precipitate may form. Mix gently before use. Store at 2-8°C until ready to use or expiry.
3. 100× Conjugate for Chagas: One vial, 150 μL. This contains horseradish peroxidase-labeled anti-human IgG antibody. Mix well prior to use. The 100× Conjugate is added to the Conjugate Diluent before use. Store at 2-8°C until expiry.
4. Conjugate Diluent for Chagas Detect: One bottle, 12 mL, is to be used to dilute the 100× conjugate before adding to the ELISA plate. Stable at 2-8°C until the expiration date.
5. Chagas Negative Control: One vial, 50 μL of heatinactivated serum. The negative control will aid in monitoring the integrity of the kit. Store at 2-8°C until expiry.
6. Chagas Positive Control: One vial, 50 μL of heatinactivated diluted serum. The positive control will aid in monitoring the integrity of the kit. Store at 2-8°C until expiry.
7. Chagas Cut-off Control: One vial, 50 μL of heatinactivated diluted serum. The positive control will aid in monitoring the integrity of the kit. Store at 2-8°C until expiry.
8. 10× Wash Buffer: One bottle, 120 mL of Wash Buffer to be used for all the wash steps of this procedure. Store at 2-8°C until ready to use or expiry.
9. Liquid TMB Substrate: One bottle, 12 mL, ready-to-use. Store at 2-8°C until ready to use or expiry. The substrate
should be kept in a light-protected bottle at all times as provided.
10. Stop Solution: One bottle, 6 mL, to be used to stop the reaction. Store at 2-8°C until ready to use or expiry
NOTE: All reagents and controls must be allowed to reach room temperature (~25°C) and mixed thoroughly by gentle inversion prior to use. Always practice sterile and aseptic techniques at every step. For example, open all reagents in a BSL-2 hood to avoid contamination with airborne bacteria to maintain shelf life.
