Intended Use

This assay employs a quantitative enzyme immunoassay technique that measures the Plasminogen Activator Inhibitor-1 (PAI-1) in samples.

Contents of Kit

1. Human PAI-1 Microplate: A 96-well polystyrene microplate (12 strips of 8 wells) coated with a polyclonal antibody against human PAI-1.
2. Sealing Tapes: Each kit contains 3 precut, pressure sensitive sealing tapes that can be cut to fit the format of the individual assay.
3. Human PAI-1 Standard: Human PAI-1 in a buffered protein base (2.25 ng, lyophilized).
4. Biotinylated Human PAI-1 Antibody (50×): A 50-fold concentrated biotinylated polyclonal antibody against human PAI-1 (120 μl).
5. MIX Diluent Concentrate (10×): A 10-fold concentrated buffered protein base (30 ml).
6. Standard Diluent (1×): A buffered protein base with stabilizer (2 ml).
7. Wash Buffer Concentrate (20×): A 20-fold concentrated buffered surfactant (30 ml, 2 bottles).
8. SP Conjugate (100×): A 100-fold concentrate (80 μl).
9. Chromogen Substrate (1×): A stabilized peroxidase chromogen substrate tetramethylbenzidine (7 ml).
10. Stop Solution (1×): A 0.5 N hydrochloric acid solution to stop the chromogen substrate reaction (11 ml).

Storage

1. Upon arrival, immediately store components of the kit at recommended temperatures up to the expiration date.
2. Store Standard, SP Conjugate, and Biotinylated Antibody at -20°C.
3. Store Microplate, Diluent Concentrate (10×), Standard Diluent (1×), Wash Buffer, Stop Solution, and Chromogen Substrate at 2-8°C.
4. Unused microplate wells may be returned to the foil pouch with the desiccant packs and resealed. May be stored for up to 30 days in a vacuum desiccator.

Performance Characteristics

1. This assay recognizes both natural and recombinant human PAI-1.
2. The minimum detectable dose of human PAI-1 as calculated by 2SD from the mean of a zero standard was established to be 14 pg/ml.
3. Intra-assay precision was determined by testing three plasma samples twenty times in one assay.
4. Inter-assay precision was determined by testing three plasma samples in twenty assays.

General Description

Type I plasminogen activator inhibitor (PAI-1) is a 50 kDa serpin family member that inhibits tissue- and urokinase-type plaminogen activators (tPA, uPA). This protein appears to be an important regulator of plasminogen activation by tPA and extracellular proteolysis by uPA. The plasminogen activator proteolytic enzyme systems are important not only for fibrinolysis but also for extracellular matrix remodeling and have been implicated in a number of normal and pathological processes including angiogensis, ovulation and embryogenesis, thrombotic and hemorrhagic disorders, connective tissue diseases, neoplasm, and sepsis. PAI-1 is a prognosticator in breast cancer, gastric cancer, various forms of lung cancer and cervical cancer.
