Background
Potato is an important food and cash crop, and its market demand and planting area are also increasing. With the selection and optimization of varieties, the yield and quality of potato have been significantly improved, but the consequent occurrence of pests and diseases is also becoming more and more serious. Among them, potato virus disease is a kind of disease that often appears in the process of potato growth, and it is the most common, the most important and the most harmful kind of disease that occurs in potato, which will seriously affect the healthy growth of potato, and thus affect the yield and quality of potato, leading to the decline of economic benefits.
Potato leafroll disease is caused by Potato leafroll virus (PLRV), which is mainly transmitted by aphids. Typical symptoms are upward curling of the leaf margins centered on the main vein, and in the early stages of the disease, the symptoms consist mainly of fading green of the plant's top young leaves, which then curl upward along the central vein to the older leaves. In severe cases, the disease forms a cylindrical shape, the color of the leaf blade becomes pale and, in some cases, slightly purplish, the leaf blade becomes small, thick and brittle, and the leaf veins become hard. Diseased plants also undergo varying degrees of dwarfing, with black spots in cross sections of the stems, more pronounced disease at the base of the stems and at the nodes, and tuber tissues that exhibit reticulate necrotic mottling in the ductal area.
PLRV belongs to the genus Polerovirus in the family Solemoviridae, and the viral particles are isometric, with a diameter of 24 nm, and contain a single-stranded positive-sense RNA genome of about 5.9 kb. Polerovirus does not have a 3' poly(A) tail and the open reading frames (including those encoding structural proteins) are aligned in the genome. The viral capsid is icosahedral and consists of capsid protein (CP) and a small amount of read-through protein (RTP) that reads the CP termination codon for translation through the ribosome. The read-through domain (RTD), located in the RTP portion after the CP stop codon, regulates aphid dispersal along with motifs in the CP. The immune response of PLRV-infected aphids to the aphid-infecting virus, Myzus persicae densovirus (MpDNV), was suppressed, resulting in elevated titers of MpDNV in aphids that acquired PLRV virus. In the absence of replicating PLRV, transient expression of PLRV P0 in plants was sufficient to recapitulate the increase in MpDNV titers in aphids. The P0 protein may be selected to manipulate the response of aphids to densovirus infection, in part to induce aphids to form alates, thereby facilitating the spread of both viruses in crops.
