This product (Catalog No. CABT-BL1344) is a rabbit-derived polyclonal antibody raised against human ECD, targeting the amino acid region 10–406 of the protein. It has been affinity purified by Protein A. Ecdysoneless homolog (ECD) is a highly conserved, essential protein acting as a, key regulator of cell cycle progression (G1-S transition), pre-mRNA splicing, and stability of tumor suppressors like p53 and Rb. It is the human ortholog of the Drosophila Ecd protein, which is vital for metamorphosis, and its dysfunction leads to severe developmental issues. In humans, ECD is overexpressed in various cancers (e.g., breast, cervical), correlating with poor prognosis.
CABT-BL1344 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
CABT-BL1344 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.

Abstract

Enzyme-prodrug combination is a recognized therapeutic strategy for the treatment of cancer cells. Cytosine deaminases (CD) are non-mammalian proteins perceived to convert the prodrug 5-fluorocytosine (5-FC), into the active antitumor agent 5-fluorouracil (5-FU). In this study, we have synthesized 5-FC as well as 5-FU loaded chitosan-silver nanoparticles in a green synthesis approach, with high yield. The size of the nanoparticles was ascertained to be below 25 nm, thus proficient in penetrating cells. Investigation in human breast carcinoma cell line MDA-MB-468 manifested potent cytotoxicity for 5-FU nanoparticles as compared to the prodrug, 5-FC nanoparticles. E. coli CD (ECD) effectively hydrolyzes 5-FC into 5-FU but was inert to silver nanoparticles as well as 5-FU loaded nanoparticles.

Sex dimorphic adaptive responses against Azadirachtin toxicity in gut tissues ofSpathosternum prasiniferum prasiniferum(Orthoptera; Acridoidea)

Abstract

Acridids (short-horned-grasshopper) are distributed largely in the agricultural-field, grass-lands, and forests. It may act as a dependable bio-indicator species in response to ecological-stress. Among the acridids found in West Bengal,Spathosternum prasiniferum prasiniferum(Walker, 1871) are multivoltine in nature and were considered for the present study. Azadirachtin is a tetranortri terpenoids present in neemtree (Azadirachta indica),which develops antifeedancy/ growth-regulation/ fecundity-suppression/ sterilization/ oviposition/ repellence and deformity in insect via oxidative stress by the production of free radicals and causes their death. Antioxidants protect insects by scavenging free radicals. This study explored Azadirachtin toxicity on food-indices/biochemical/physiological influences on mid-gut ofSpathosternum prasiniferum prasiniferum.Commercially available Azadirachtin was applied (1-20 ppm) onS. pr. prasiniferumfor in vivo/in vitro experiment, where gut-tissues were analysed to determine oxidative-stress-markers (ALP/MDA/NPSH/AChE), antioxidant-markers (SOD/CAT/GPx). Food-weight/insect-weight/excreta-weight were recorded before/after the experiment to evaluate the consumption-index(CI), approximate-digestibility(AD), efficiency of conversion of digested-food(ECD) and efficiency of conversion of ingested-food(ECI). Nutrient metabolizing-enzymes (cellulase/amylase) were screened. Azadirachtin (>7 ppm) decreased ECD and restricted food-consumption that increased insect-mortality (50-80%). The gut MDA significantly increased with an impairment of soluble-thiols. The antioxidant-enzymes were variably impaired resulting in tissue-damages more in male than female. Finally, Azadirachtin influenced nutrient-metabolizing-enzymes and antioxidant-enzymes indicating these parameters to be good stress-markers. This finding might be extrapolated for further evaluation of the ecological impact of Azadirachtin on the food chain/web in a composite-ecosystem.
