This product (Catalog No. CABT-BL562) is a rabbit-derived polyclonal antibody raised against human C15orf39, targeting the amino acid region 928–1047 of the protein. It has been affinity purified by Protein A. C15orf39 is a protein-coding gene located on the long arm of human chromosome 15 at cytogenetic band q24.2, spanning genomic coordinates 75,198,889 to 75,212,169 (GRCh38 assembly), and it encodes the uncharacterized protein C15orf39, a member of the Atrophin-1 family with five exons and multiple isoforms. The protein C15orf39 is primarily expressed in a broad range of tissues, with notably high levels in the testis (RPKM 35.8) and bone marrow (RPKM 21.8), as well as moderate expression in fetal tissues such as adrenal, heart, intestine, kidney, lung, and stomach during gestational weeks 10-20. Functionally, C15orf39 acts as a negative regulator of inflammatory responses in human microglial cells, where it localizes to both the cytosol and nucleus; it is phosphorylated by mitogen-activated protein kinase 1 (MAPK1) at serine 364 and interacts with proteins such as MAD2L1 and protein arginine methyltransferase 2 (PRMT2). In lipopolysaccharide (LPS) and interferon-gamma (IFNG)-stimulated microglial HMC3 cells, C15orf39 overexpression suppresses the expression of pro-inflammatory cytokines including interleukin-6 (IL6) and tumor necrosis factor-alpha (TNFA), while its knockdown enhances cytokine production; this regulation occurs through cytoplasmic interaction with PRMT2 to inhibit nuclear factor kappa B (NFKB) activation, forming a negative feedback loop that limits microglia-mediated inflammation.
CABT-BL562 is suitable for use in Western blot (WB) (1:500–1:1,000) application.
CABT-BL562 is supplied as a liquid formulation in PBS with 0.02% sodium azide and 50% glycerol, pH 7.3. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
