This product (Catalog No. CABT-BL506) is a rabbit-derived polyclonal antibody raised against human C11orf53. This antibody exhibits cross-reactivity with C11orf53 from mouse and rat. It has been affinity purified by Protein A. C11orf53, officially designated as POU2AF2 (POU class 2 homeobox associating factor 2), is a protein-coding gene located on the long arm of human chromosome 11 at cytogenetic band 11q23.1, spanning genomic coordinates 111245725 to 111286401 on reference sequence NC_000011.10. It encodes a nuclear protein that functions as a transcriptional coactivator, enabling sequence-specific DNA binding and positive regulation of DNA-templated transcription, with aliases including OCAT1 and OCA-T1. The protein product of POU2AF2, often referred to interchangeably as C11orf53 in early literature, directly interacts with the transcription factor POU2F3 to form a heterodimer that maintains chromatin accessibility and enhancer activity at target genes. This interaction is critical for the development and identity of tuft cells, rare chemosensory epithelial cells that coordinate immune and neural responses in mucosal tissues. In the context of oncology, POU2AF2 is highly coexpressed and selectively essential in the tuft cell-like subtype of small cell lung cancer (SCLC-P), where it drives enhancer H3K27 acetylation, supports POU2F3-dependent gene expression (including pathways like E2F, G2M, and MYC), and promotes tumor cell viability, proliferation, and xenograft growth. Depletion of POU2AF2 via CRISPR-Cas9 reduces chromatin accessibility, induces apoptosis, and impairs tumor progression specifically in SCLC-P cells, without affecting other SCLC subtypes (A, N, Y), highlighting its potential as a therapeutic target for subtype-specific interventions.
