This product (Catalog No. DPABH-24686) is a rabbit-derived polyclonal antibody raised against human TPP1, targeting the amino acid region 1–320 of the protein. This antibody exhibits cross-reactivity with Human TPP1 from mouse and rat. It has been affinity purified by Protein A. Tripeptidyl-peptidase I (TPP I), encoded by the TPP1 gene, is a lysosomal enzyme synthesized as an inactive proenzyme that undergoes proteolytic maturation in acidic lysosomal conditions to become active. It primarily exhibits aminopeptidase activity with minor endopeptidase function, using Ser475 as the active-site nucleophile to hydrolyze unmodified N-termini of small, unstructured polypeptides. TPP I is essential for lysosomal protein catabolism, digesting peptides into tripeptides for recycling, and its deficiency due to TPP1 mutations causes neuronal ceroid lipofuscinosis type 2 (CLN2 disease), a fatal neurodegenerative lysosomal storage disorder characterized by accumulation of undigested material in neurons, leading to seizures, vision loss, and motor impairment starting in early childhood.
DPABH-24686 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-24686 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
