General Notes
This product (Catalog No. DPABH-15112) is a rabbit-derived polyclonal antibody raised against human TMBIM6, targeting the amino acid region 1–86 of the protein. This antibody exhibits cross-reactivity with Human TMBIM6 from mouse. It has been affinity purified by Protein A. TEGT, or Testis Enhanced Gene Transcript (also known as TMBIM6), is a human gene located on chromosome 12q13.12 that encodes the protein known as Bax inhibitor 1 (BI-1), an evolutionarily conserved endoplasmic reticulum (ER)-resident transmembrane protein with six predicted transmembrane domains. This protein functions primarily as an anti-apoptotic regulator, inhibiting cell death pathways controlled by BCL2 family members such as BAX, while interacting with anti-apoptotic proteins like BCL2 and BCL-XL but not pro-apoptotic BAX or BAK. Overexpression of BI-1 suppresses apoptosis induced by various stressors, including etoposide, staurosporine, growth factor deprivation, and ER stress, whereas its knockdown or knockout heightens cellular sensitivity to ischemia-reperfusion injury and ER stress in tissues like the liver and kidney. Originally identified in rat testis as a developmentally regulated, single-copy gene with testis-enriched expression, the human homolog was cloned in 1995 and later recognized for its role in cytoprotection; initially not assigned to any known vertebrate gene family, it is now classified as part of the TMBIM (Transmembrane BAX Inhibitor Motif-containing) family. BI-1's protective effects extend to modulating ER stress responses, preventing activation of pathways like the unfolded protein response, and it has been implicated in diseases involving excessive apoptosis, such as ischemia-related organ damage. The gene produces multiple transcripts via alternative polyadenylation, with the protein featuring cytosolic N- and C-termini and no significant homology to other eukaryotic proteins beyond distant bacterial sequences.
DPABH-15112 is suitable for use in Western blot (WB) (1:500–1:2,000), IP (0.5-4.0 μg for 1.0-3.0 mg of total protein lysate), IHC (1:50–1:500) applications.
DPABH-15112 is supplied as a liquid formulation in PBS with 0.02% sodium azide and 50% glycerol, pH 7.3. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
