General Notes
This product (Catalog No. DPABH-10322) is a rabbit-derived polyclonal antibody raised against human SKAP2, targeting the amino acid region 111–222 of the protein. It has been affinity purified by Protein A. SKAP2 is a protein-coding gene located on the short arm of human chromosome 7 at position 7p15.2, encoding src kinase-associated phosphoprotein 2 (SKAP2), an adaptor protein that functions as a substrate for Src family kinases and plays a critical role in signal transduction pathways, particularly those regulating immune cell activation, adhesion, and cytoskeletal dynamics. The SKAP2 protein, also known as SKAP55R or RA70, shares homology with SKAP1 (SKAP55) and features a modular structure including an N-terminal coiled-coil domain that enables self-dimerization, a central pleckstrin homology (PH) domain for binding membrane lipids, and a C-terminal Src homology 3 (SH3) domain that facilitates interactions with proline-rich proteins such as FYB (SLAP-130). It is phosphorylated by kinases like FYN but not LCK or ZAP70, and it links upstream receptors—such as the B-cell receptor (BCR) or CD11b/CD18 integrin—to downstream effectors, thereby coupling antigen recognition to cytoskeletal reorganization and cell motility in B cells, macrophages, and neutrophils. Additionally, SKAP2 inhibits actin polymerization by interacting with assembly factors like WAVE2 and ARP2/3, which may suppress tumor cell invasiveness and modulate inflammation through pathways like TLR4-NF-κB.
DPABH-10322 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-10322 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
