General Notes
This product (Catalog No. DPABH-18833) is a rabbit-derived polyclonal antibody raised against human RWDD2B. This antibody exhibits cross-reactivity with RWDD2B from mouse. It has been affinity purified by Protein A. RWDD2B is a protein-coding gene in humans that encodes RWD domain-containing protein 2B, a small protein featuring a conserved RWD domain involved in protein-protein interactions and a domain of unknown function (DUF1115). Located on the long arm of chromosome 21 at cytogenetic band 21q21.3, spanning approximately 15 kb with six exons, RWDD2B produces two main isoforms via alternative splicing, with the longer isoform consisting of 319 amino acids. Its expression is ubiquitous across human tissues, with the highest levels observed in the testis (RPKM 9.4) and prostate (RPKM 8.5), and it is also detectable in fetal tissues such as adrenal, heart, and kidney during the second trimester of gestation. Although the precise molecular function of RWDD2B remains incompletely understood, it has been associated with several notable biological contexts. The gene maps to a critical region on chromosome 21 linked to the monosomy 21 phenotype, and its murine ortholog (orf5) shares this positional relevance. Epigenetic and gene expression studies have identified RWDD2B as a target of osteoarthritis (OA) genetic risk, where methylation quantitative trait loci (meQTLs) near the gene influence its expression in joint tissues, potentially contributing to disease susceptibility through regulatory mechanisms responsive to histone deacetylase inhibitors like trichostatin A (TSA). Additionally, protein interaction data indicate that RWDD2B binds to BRCA1, suggesting a potential role in the cellular response to transcription-associated DNA damage, though this interaction was detected via two-hybrid screening and requires further validation. The gene's distal promoter contains cis-regulatory elements sensitive to TSA and serum stimulation, highlighting its involvement in transcriptional regulation. Aliases for RWDD2B include GL011 and C21orf6, reflecting its historical nomenclature as chromosome 21 open reading frame 6.
