General Notes
This product (Catalog No. DPABH-02542) is a rabbit-derived polyclonal antibody raised against human OR6N1. This antibody exhibits cross-reactivity with OR6N1 from mouse and rat. It has been affinity purified by Protein A. OR6N1 is a protein-coding gene in humans that encodes the olfactory receptor protein OR6N1, a G protein-coupled receptor (GPCR) with seven transmembrane domains responsible for detecting odorant molecules in the nasal epithelium to initiate a neuronal response and trigger the perception of smell. As part of the largest gene superfamily in the human genome, consisting of approximately 391 intact olfactory receptor genes and around 465 pseudogenes, part of over 850 total loci clustered across 51 sites on 21 chromosomes, OR6N1 belongs to subfamily 6N and is thought to recognize one or more structurally related odorants, contributing to the combinatorial coding of olfactory signals. The OR6N1 gene is located on the reverse strand of chromosome 1 at position 1q23.1 (GRCh38 coordinates: 158,747,814–158,772,195), spanning about 24.4 kb and containing a single coding exon that produces a 312-amino-acid protein with a molecular mass of 34,869 Da. Structurally, the encoded protein features typical GPCR motifs, including glycosylation sites and a predicted high-confidence 3D structure via AlphaFold modeling, enabling ligand binding and signal transduction through G protein activation. Expression of OR6N1 is primarily restricted to olfactory receptor neurons in the nose, though database analyses indicate low-level presence in tissues such as bone marrow and granulocytes, potentially hinting at non-olfactory roles that remain uncharacterized. While specific odorant ligands for OR6N1 have not been definitively identified, olfactory receptors like it are screened in heterologous systems against panels of odorants to map responsiveness, underscoring their role in sensory perception. Genetic variants in OR6N1, including missense mutations of uncertain significance (e.g., p.Phe68Tyr), show high intolerance to variation, suggesting potential functional constraints, but no direct associations with human diseases have been firmly established beyond tentative links in genome-wide studies. Evolutionarily, OR6N1 has orthologs in mammals like chimpanzees (98% similarity) and mice (85%), tracing back to chordate ancestors, and it interacts with over 100 other proteins, primarily fellow olfactory receptors, in predicted networks.
DPABH-02542 is suitable for use in WB 1 (1000-3000), IHC 1 (50-1:200) applications.
