General Notes
This product (Catalog No. DPABH-21681) is a rabbit-derived polyclonal antibody raised against human OR2T11. It has been affinity purified by Protein A. OR2T11 is a human gene that encodes the olfactory receptor 2T11 (OR2T11), a G protein-coupled receptor (GPCR) belonging to the olfactory receptor family 2 subfamily T, which plays a key role in detecting odorant molecules to initiate the perception of smell. This receptor specifically responds to low-molecular-weight thiols, such as ethyl mercaptan and tert-butyl mercaptan, enabling the ultrasensitive detection of volatile sulfur compounds associated with environmental threats like rancid food or predators. Notably, OR2T11's activation by these thiols is regulated by ionic copper, marking the first identified instance of a metal ion's involvement in human olfaction, with silver ions and nanoparticles mimicking this effect. The OR2T11 gene is located on the long arm of chromosome 1 at position 1q44 and consists of two exons, producing a protein with seven transmembrane domains typical of GPCRs. As part of the largest gene family in the human genome, olfactory receptors like OR2T11 share a structure that facilitates odorant recognition and signal transduction through G protein mediation. However, OR2T11 is a segregating pseudogene, meaning genetic polymorphisms result in alleles that are either functional (encoding a full protein) or non-functional (predicted to produce no viable receptor) across individuals, contributing to variation in olfactory repertoires. Research on OR2T11 has highlighted its specificity for short-chain thiols (one to five carbons) while showing no response to similar alcohols, underscoring the receptor's compact binding pocket and the profound perceptual differences in sulfur versus non-sulfur odors. Studies employing techniques like site-directed mutagenesis, NMR spectroscopy, and computational modeling have elucidated these mechanisms, revealing potential implications for understanding metal homeostasis in sensory biology.
