This product (Catalog No. DPABH-19322) is a rabbit-derived polyclonal antibody raised against human LRRC3B. This antibody exhibits cross-reactivity with LRRC3B from mouse. It has been affinity purified by Protein A. LRRC3B (leucine-rich repeat containing 3B) is a human gene located on the short arm of chromosome 3 at position 3p24.1, encoding a 259-amino-acid protein characterized by leucine-rich repeat domains that function as a tumor suppressor by inhibiting cell proliferation and replication in epithelial cells. The LRRC3B protein contains conserved structural motifs, including an N-terminal leucine-rich repeat domain (LRRNT) and multiple leucine-rich repeat motifs (LRR), which are implicated in protein-protein interactions essential for its regulatory roles. Expression of LRRC3B is biased toward the brain (RPKM 1.4) and testis (RPKM 0.6), with detectable levels in various fetal tissues such as adrenal, heart, intestine, kidney, lung, and stomach during 10–20 weeks of gestation. In biological processes, LRRC3B confers resistance to ultraviolet (UV) radiation-induced damage and accelerates DNA repair rates in cells, as demonstrated by overexpression studies in HeLa cells that also downregulate cyclin B1 levels to suppress proliferation. The gene's promoter is regulated by DNA methylation; hypermethylation leads to transcriptional silencing, a mechanism reversible by demethylating agents. This epigenetic inactivation is frequently observed in multiple epithelial malignancies, including gastric, renal, colorectal, lung, and breast cancers, where LRRC3B expression is significantly downregulated compared to normal tissues. In non-small cell lung cancer (NSCLC), reduced LRRC3B levels correlate with increased cell proliferation and invasion, highlighting its role in tumor suppression. Moreover, LRRC3B expression and promoter hypomethylation status serve as predictive biomarkers for response to anti-PD-1 immunotherapy, influencing the anti-tumor immune microenvironment through mechanisms like lymphocyte infiltration. These findings underscore LRRC3B's potential as a therapeutic target in cancer treatment.

DPABH-19322 is supplied as a liquid formulation in Phosphate buffered saline, pH 7.4, 150mM NaCl, 0.02% sodium azide and 50% glycerol. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
