This product (Catalog No. DPABH-05466) is a rabbit-derived polyclonal antibody raised against human LINGO2. This antibody exhibits cross-reactivity with LINGO2 from mouse and rat. It has been affinity purified by Protein A. LINGO2, officially known as leucine rich repeat and Ig domain containing 2, is a protein-coding gene in humans that encodes a transmembrane protein with leucine-rich repeat (LRR) domains, an immunoglobulin-like domain, and a cytoplasmic tail, predicted to play a role in the positive regulation of synapse assembly. The gene is located on the short arm of chromosome 9 at position 9p21.2-p21.1 and consists of 20 exons, producing a 606-amino acid protein that is structurally similar to LINGO1, sharing about 60% amino acid identity. LINGO2 is predicted to localize to the cell membrane and synaptic structures, including glutamatergic synapses, where it may influence neuronal connectivity and function. Research has linked genetic variants in LINGO2 to several neurological and metabolic conditions, highlighting its potential role in disease pathogenesis. Specifically, polymorphisms in LINGO2 have been associated with an increased risk of essential tremor, a common movement disorder, and Parkinson's disease, suggesting involvement in motor control pathways. In Asian populations, the LINGO2 variant rs7033345 has been associated with increased risk of Parkinson's disease. Beyond neurology, LINGO2 variants contribute to gestational diabetes mellitus risk in Chinese populations and are linked to body mass variations in elderly individuals, indicating broader regulatory effects on metabolism and stem cell phenotypes. Additionally, LINGO2 expression has been implicated as a marker in gastric cancer stem cells, correlating with tumor progression, motility, and patient outcomes. Expression of LINGO2 is generally low across human tissues, with minimal levels observed in fetal samples from organs such as the adrenal gland, heart, and lung during early development (10-20 weeks gestation). The protein's conserved domains, including multiple LRR motifs and an I-set immunoglobulin domain, underscore its evolutionary role in cell-cell interactions, particularly in the nervous system. DPABH-05466 is supplied as a liquid formulation in Phosphate buffered saline, pH 7.4, 150mM NaCl, 0.02% sodium azide and 50% glycerol. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
