This product (Catalog No. DPABH-25747) is a rabbit-derived polyclonal antibody raised against human GCK, targeting the amino acid region 1–465 of the protein. It has been affinity purified by Protein A. GCK functions as a critical glucose sensor in pancreatic beta cells and hepatocytes, catalyzing the initial step of glycolysis by phosphorylating glucose to glucose-6-phosphate. Unlike other hexokinases, GCK possesses a low affinity for glucose, allowing its activity to scale linearly with physiological blood sugar levels. This unique kinetic profile makes it the primary regulator of insulin secretion and hepatic glycogen synthesis. Mutations in the GCK gene are clinically linked to Maturity-Onset Diabetes of the Young (MODY2) and persistent hyperinsulinemic hypoglycemia. Consequently, GCK-targeted antibodies are vital for studying glucose homeostasis and developing small-molecule activators for type 2 diabetes therapy.
DPABH-25747 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-25747 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
