This product (Catalog No. DPABH-09144) is a rabbit-derived polyclonal antibody raised against human ERLIN1, targeting the amino acid region 29–173 of the protein. It has been affinity purified by Protein A. ERLIN1 is a protein-coding gene located on chromosome 10q24.31 that encodes erlin-1, a 348-amino acid integral membrane glycoprotein with a molecular mass of approximately 39 kDa, belonging to the Stomatin-prohibitin-flotillin-HflC/K (SPFH) superfamily due to its conserved prohibitin (PHB) domain. Erlin-1 primarily localizes to the endoplasmic reticulum (ER) and nuclear envelope, where it forms heterooligomeric complexes with the related protein erlin-2, defining detergent-resistant, lipid raft-like microdomains enriched in cholesterol and sphingolipids. These complexes function as scaffolds for ER-associated protein degradation (ERAD), facilitating the ubiquitination and proteasomal degradation of substrates such as activated inositol 1,4,5-trisphosphate receptors (IP3Rs) and 3-hydroxy-3-methylglutaryl-CoA reductase (HMGCR), thereby regulating calcium signaling and sterol biosynthesis. Additionally, erlin-1 binds cholesterol and interacts with the SREBP-SCAP-INSIG pathway to maintain cellular cholesterol homeostasis by inhibiting SREBP-2 activation under high sterol conditions.
DPABH-09144 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-09144 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
