This product (Catalog No. DPABH-28249) is a rabbit-derived polyclonal antibody raised against human EEPD1. It has been affinity purified by Protein A. Endonuclease/exonuclease/phosphatase family domain containing 1 (EEPD1) is a protein encoded by the EEPD1 gene in humans, located on chromosome 7p14.2, that functions as a DNA 5'-endonuclease essential for promoting 5' end resection at double-strand breaks occurring at stalled replication forks, thereby facilitating DNA repair through homologous recombination and maintaining genome stability. The EEPD1 protein consists of approximately 569-573 amino acids and features two N-terminal helix-hairpin-helix DNA-binding domains, a C-terminal DNase I-like domain, and a nuclease active site that spans the N- and C-terminal regions, enabling its endonuclease activity in a magnesium-dependent manner. EEPD1 is rapidly recruited to sites of replication stress, where it interacts with proteins such as EXO1, RPA32, and BLM to initiate repair processes, while also inhibiting alternative pathways like classical nonhomologous end joining and microhomology-mediated end joining. Depletion of EEPD1 leads to accumulation of cells in S and G2 phases, reduced proliferation, and increased genome instability, highlighting its critical role in replication fork restart and downstream activation of DNA damage response pathways involving ATR/CHK1 and H2AX phosphorylation.
DPABH-28249 is suitable for use in Western blot (WB) (1:200–1:1,000), IF (immunofluorescence) (1:20–1:100) applications.
DPABH-28249 is supplied as a liquid formulation in pH7.4 PBS, 0.05% sodium azide, 40% Glycerol. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
