General Notes
This product (Catalog No. DPABH-28537) is a rabbit-derived polyclonal antibody raised against human CNR1, targeting the amino acid region 2–116 of the protein. It has been affinity purified by Protein A. The cannabinoid receptor 1 (CB1) is a G protein-coupled receptor (GPCR) that serves as the principal molecular target for endocannabinoids in the central nervous system (CNS), where it modulates neurotransmitter release and synaptic plasticity to regulate diverse physiological processes such as pain perception, appetite, mood, and memory. Structurally, CB1 features seven transmembrane α-helices typical of class A GPCRs, with a characteristic "twin toggle switch" involving residues F2003.36 and W3566.48 that facilitate conformational changes upon ligand binding; high-resolution crystal structures, including those bound to antagonists like AM6538 (PDB: 5TGZ) and agonists like AM11542 (PDB: 5XRA), have revealed an extracellular vestibule for ligand access and an allosteric modulatory pocket. CB1 can form homodimers or heterodimers with other GPCRs, influencing its signaling dynamics. CB1 is predominantly expressed in the CNS, with high densities in the neocortex, hippocampus, basal ganglia, cerebellum, and brainstem, as well as moderate levels in peripheral tissues such as adipose tissue, liver, and testis; this widespread distribution underscores its role in both neural and non-neural functions.
DPABH-28537 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-28537 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
