This product (Catalog No. DPABH-04479) is a rabbit-derived polyclonal antibody raised against human CDK5RAP1, targeting the amino acid region 239–587 of the protein. This antibody exhibits cross-reactivity with Human CDK5RAP1 from mouse and rat. It has been affinity purified by Protein A. CDK5RAP1, also known as CDK5 regulatory subunit-associated protein 1 or C42, is a human gene located on chromosome 20q11.21 that encodes a multifunctional protein serving as both an inhibitor of cyclin-dependent kinase 5 (CDK5) and a radical S-adenosylmethionine (SAM) enzyme acting as a methylthiotransferase for RNA. The protein represses CDK5 activity by preventing the formation of its active complex with regulatory subunits like p35, thereby modulating neuronal differentiation, neurocytoskeleton dynamics, and processes implicated in neurodegeneration. In its enzymatic role, CDK5RAP1 specifically catalyzes the conversion of N⁶-isopentenyladenosine (i⁶A) to 2-methylthio-N⁶-isopentenyladenosine (ms²i⁶A) at position 37 in the anticodon loops of four mitochondrial tRNAs (for phenylalanine, tryptophan, tyrosine, and serine UCN), a modification essential for accurate mitochondrial translation, codon recognition, and anticodon-codon stability. This activity extends beyond mitochondria, as the protein also introduces ms²i⁶A into nuclear RNAs, including polyadenylated RNAs and small RNAs like miRNAs, potentially influencing RNA structure, processing, and function. Alternative splicing generates multiple isoforms, with variant 1 featuring an N-terminal mitochondrial targeting signal for localization in mitochondria, while variant 2 resides in the cytosol and nucleus, enabling compartmental-specific functions. Expression of CDK5RAP1 is ubiquitous across human tissues, with elevated levels in high-mitochondrial-activity organs like muscle and brain.

DPABH-04479 is supplied as a liquid formulation in PBS with 0.02% sodium azide and 50% glycerol, pH 7.3. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
