SNRNP70 Introduction
Introduction
The SNRNP70 (small nuclear ribonucleoprotein U1 subunit 70) gene encodes a core protein component of the U1 small nuclear ribonucleoprotein (U1 snRNP), which is essential for pre-mRNA splicing. As the largest subunit of the U1 snRNP complex, SNRNP70 plays a critical role in recognizing the 5' splice site during the earliest steps of spliceosome assembly. Beyond its canonical function in RNA processing, this protein has emerged as a key player in protecting the transcriptome from premature cleavage and polyadenylation, and its dysfunction is increasingly linked to human diseases ranging from autoimmune disorders to cancer and neurodegeneration. The protein is also a major autoantigen recognized by sera from patients with mixed connective tissue disease and systemic lupus erythematosus, cementing its clinical significance.

Role in Cellular Signaling and Stress Responses
SNRNP70 expression and activity are modulated by various cellular signals. The gene is regulated by dexamethasone, hydrogen peroxide, ascorbic acid, TGFB1, and CTNNB1, suggesting integration into broader signaling networks. In rat cells, Snrnp70 is involved in cellular responses to retinoic acid, transforming growth factor beta, and tumor necrosis factor, indicating that splicing factor regulation is part of the cellular response to extracellular cues. These findings link core splicing machinery to signal transduction pathways that control cell proliferation, differentiation, and inflammation.
Conclusion
SNRNP70 encodes a multifunctional RNA-binding protein that stands at the center of eukaryotic gene expression. As the largest subunit of the U1 snRNP, it orchestrates 5' splice site recognition, initiates spliceosome assembly, and protects the transcriptome from premature cleavage and polyadenylation. Its recently discovered role in tethering lncRNAs to chromatin expands our understanding of how noncoding RNAs are regulated and localized. Clinically, SNRNP70 spans the spectrum from autoimmunity to oncology and neurodegeneration: it is a major autoantigen in mixed connective tissue disease and SLE , an emerging prognostic biomarker and potential oncogene in hepatocellular carcinoma , and a candidate player in Alzheimer's disease and ALS. The convergence of these diverse roles—fundamental RNA processing, autoimmune targeting, and cancer progression—positions SNRNP70 as a fascinating subject for continued research and a promising therapeutic target across multiple disease areas.
