netrin 4
NTN4 Introduction
NTN4 encodes netrin-4 (also known as beta-netrin), a secreted protein of the netrin family that guides the behavior of cells during the development of the nervous system, blood vessels, and other tissues. Netrins were first characterized as axon guidance cues: they are secreted by target cells and steer growing axons by attracting or repelling them through receptors of the DCC and UNC5 families. Netrin-4 shares the laminin-like domain architecture of its relatives, but it has evolved distinctive properties, most notably an unusually strong ability to bind the gamma chains of laminins and to incorporate into basement membranes, the sheet-like extracellular matrices that underlie epithelia and surround blood vessels. Through this association, netrin-4 influences the assembly and function of basement membranes and thereby affects angiogenesis, vascular permeability, and the behavior of epithelial cells. In the nervous system, netrin-4 contributes to axon guidance, neuronal migration, and the differentiation of oligodendrocyte precursor cells. The protein also has a well-documented role in cancer, where its effects are context-dependent: in some tumor types netrin-4 suppresses growth and metastasis by modulating the tumor microenvironment, whereas in others it has been reported to support tumor progression. This duality has made netrin-4 a subject of active investigation in vascular biology and oncology.

Secreted Protein Architecture and Laminin-Binding Properties
Netrin-4 is a secreted glycoprotein whose domain organization includes an N-terminal laminin VI-like domain, laminin V-like (EGF-repeat) domains, and a C-terminal netrin (C345C/NTR) module.
This architecture is shared with the other netrins and, more distantly, with components of the extracellular matrix, reflecting the evolutionary origin of the family.
The NTR domain, together with the laminin-like repeats, mediates binding to the gamma-1 and gamma-3 chains of laminins, an interaction that anchors netrin-4 within basement membranes.
The NTN4 gene is located on human chromosome 12, and the protein is expressed in the developing and adult nervous system, in vascular basement membranes, and in a variety of epithelial tissues.
Unlike classical axon guidance netrins such as netrin-1, netrin-4 binds the DCC and UNC5 receptors only weakly in many assays, and much of its activity appears to be mediated through its extracellular matrix interactions.
Within basement membranes, netrin-4 can influence the polymerization of laminin networks and thereby modify the physical and signaling properties of the matrix.
The protein is subject to proteolytic processing and exists in multiple forms whose biological activities differ.
Angiogenesis, Neural Development, and Cancer Biology
In the vascular system, netrin-4 is deposited in the basement membranes of blood vessels, where it modulates endothelial cell behavior and influences vessel branching, stability, and permeability.
Studies in the developing retina and other vascular beds have shown that netrin-4 can either promote or inhibit vessel growth depending on the context, reflecting the balance of its effects on endothelial adhesion, migration, and survival.
In the nervous system, netrin-4 supports the migration and differentiation of oligodendrocyte precursor cells and contributes to the guidance of axons in the retina and elsewhere, while also influencing synapse formation.
The incorporation of netrin-4 into basement membranes is particularly important where vascular and neural structures meet, such as the blood-brain barrier and the retinal vasculature.
In cancer, reduced netrin-4 expression has been associated with enhanced tumor growth and metastasis in models of breast and other cancers, and reintroduction of the protein can normalize the tumor vasculature and restrain tumor progression.
Conversely, some studies report pro-tumorigenic roles of netrin-4 in specific malignancies, indicating that the outcome of netrin-4 signaling depends on tumor type, receptor expression, and the composition of the extracellular matrix.
