IL12B Introduction
Interleukin 12b (IL-12b), also known as natural killer cell stimulatory factor 2 (NKSF2) or cytotoxic lymphocyte maturation factor p40 (CLMF p40), is a crucial cytokine subunit encoded by the IL12B gene in humans. As a core component of the interleukin-12 (IL-12) family, IL-12b plays an indispensable role in bridging innate and adaptive immunity, regulating immune cell responses, and maintaining immune homeostasis. Unlike standalone cytokines, IL-12b exerts its biological effects primarily by forming heterodimeric complexes with other subunits, making it a versatile mediator in both host defense against pathogens and the pathogenesis of immune-related disorders. Its discovery and subsequent research have shed light on the intricate mechanisms of immune regulation, providing potential targets for the treatment of infectious diseases, autoimmune conditions, and cancer.

Core Immunological Function in Host Defense
Through its incorporation into IL-12 and IL-23, IL-12B is essential for mounting effective defenses against pathogens. The IL-12 (p40/p35) complex is the primary driver for the differentiation of naïve CD4+ T cells into T helper type 1 (Th1) cells. Th1 cells are characterized by their production of interferon-gamma (IFN-γ), which powerfully activates macrophages and other cellular immune mechanisms critical for combating intracellular pathogens like viruses, bacteria, and protozoa. Thus, signaling via the p40-containing IL-12 is central to cell-mediated immunity.
Signaling Pathway and Receptor Engagement
The biological activity of the IL-12B subunit is realized upon dimerization. The IL-12 heterodimer signals through a receptor complex composed of IL-12Rβ1 and IL-12Rβ2 chains, expressed on T cells and NK cells. Binding activates the JAK-STAT signaling pathway, predominantly leading to the phosphorylation and nuclear translocation of STAT4, which directs Th1 cell programming. When p40 pairs with p19 to form IL-23, this cytokine signals through a different receptor complex (IL-23R paired with IL-12Rβ1), activating primarily STAT3 and supporting the function of a distinct T cell subset, Th17 cells, involved in barrier defense and inflammation.
