HSPBAP1
HSPBAP1 Full Name
HSPB (heat shock 27kDa) associated protein 1
HSPBAP1 Introduction
HSPB (heat shock 27kDa) associated protein 1 (HSPBAP1), also known as PASS1, is a protein that interacts with the small heat shock protein HSPB1 (Hsp27), a molecular chaperone involved in cellular stress responses, apoptosis regulation, and cytoskeletal dynamics. Encoded by the HSPBAP1 gene on chromosome 3q21.1, HSPBAP1 contains a JmjC domain, a structural motif characteristic of the Jumonji family of proteins, many of which function as histone demethylases or chromatin modifiers. However, the precise molecular function of HSPBAP1 and whether it possesses intrinsic enzymatic activity remain areas of active investigation. The protein is widely expressed in human tissues, with particularly high levels in the testis.

Interaction with HSPB1 and Cellular Stress Responses
HSPBAP1 was identified through its physical interaction with HSPB1 (Hsp27), a ubiquitously expressed small heat shock protein that functions as an ATP-independent molecular chaperone, preventing the aggregation of misfolded proteins under stress conditions. The HSPBAP1-HSPB1 interaction may modulate HSPB1's chaperone activity, anti-apoptotic functions, and its regulatory effects on the actin cytoskeleton. HSPBAP1 contains a putative JmjC domain that shares homology with histone demethylases, though whether HSPBAP1 catalyzes histone demethylation remains unconfirmed. Some evidence suggests HSPBAP1 may localize to the nucleus under certain conditions and participate in chromatin-associated processes. The protein is also reported to be a target of stress-responsive kinases, with phosphorylation potentially regulating its subcellular localization and protein-protein interactions.
HSPBAP1 in Neurodegeneration, Spermatogenesis, and Amyotrophic Lateral Sclerosis
HSPBAP1 is predominantly expressed in the testis, where it may contribute to spermatogenesis through its association with HSPB1 and modulation of cellular stress responses during germ cell development. HSPBAP1 has also attracted attention for its potential involvement in amyotrophic lateral sclerosis (ALS): genomic studies have identified copy number variations and rare variants in the HSPBAP1 locus in patients with sporadic ALS. These genetic alterations may lead to altered protein expression, potentially disrupting the balance of stress-protective chaperone networks in motor neurons and increasing their vulnerability to the proteotoxic stress characteristic of ALS pathogenesis. Additionally, the HSPBAP1-HSPB1 pathway is being explored as a modulator of protein aggregation-related neurodegenerative processes. Further functional characterization of HSPBAP1 is needed to clarify its role in neuronal homeostasis and its potential as a therapeutic target for ALS and related motor neuron diseases.
