HSF2 Full Name
heat shock transcription factor 2
HSF2 Introduction
Heat shock factor 2 (HSF2) is a member of the heat shock factor family of transcription factors that, unlike its paralog HSF1, is primarily involved in developmental gene regulation rather than the acute stress response. Encoded by the HSF2 gene on chromosome 6q22.31, HSF2 shares the conserved structural organization of HSF family members (DNA-binding domain, trimerization domain, and transactivation domain) but has evolved distinct regulatory mechanisms and target gene repertoires. HSF2 is expressed in most tissues and is particularly important during embryogenesis, spermatogenesis, and central nervous system development.

HSF2-HSF1 Heterocomplex Formation and Functional Interplay
A defining feature of HSF2 biology is its ability to form heterotrimeric complexes with HSF1, creating a functional interplay between the two factors. During the stress response, HSF2 is recruited to a subset of HSF1 target gene promoters through heterocomplex formation, where it modulates HSF1-driven transcriptional output. This cooperation enables HSF2 to influence both the amplitude and duration of the heat shock response. In addition, HSF2 can independently bind HSEs and regulate gene expression in the absence of stress, particularly during development. HSF2 activity is regulated by post-translational modifications including SUMOylation (which promotes its interaction with HSF1), acetylation, and phosphorylation. The protein stability of HSF2 is also controlled by the ubiquitin-proteasome pathway, with the co-chaperone CHIP (C-terminus of Hsp70-interacting protein) playing a role in targeting HSF2 for degradation under non-stress conditions.
Roles in Brain Development, Spermatogenesis, and Disease
HSF2 is essential for normal brain development: Hsf2 knockout mice exhibit enlarged lateral ventricles, cortical thinning, and defects in neuronal migration, correlating with dysregulated expression of genes involved in cortical layer formation. In spermatogenesis, HSF2 regulates the expression of heat shock proteins and other factors required for post-meiotic germ cell maturation; male Hsf2-null mice display reduced fertility with defects in sperm head shaping and chromatin condensation. In cancer biology, HSF2 can function as either a tumor suppressor or oncogene depending on the tissue context: it suppresses prostate cancer progression through regulation of p53-independent senescence pathways, while in other tumor types aberrant HSF2 expression may support malignant phenotypes. The HSF2 locus is also implicated in 6q deletions associated with a subset of B-cell lymphomas. These diverse functions position HSF2 as a versatile regulator bridging developmental programming and disease pathogenesis.
