HPV 45 Introduction

Human Papillomavirus Type 45 (HPV-45) is classified as a high-risk HPV type and contributes significantly to cervical cancer globally. Although less prevalent than HPV-16 and HPV-18, HPV-45 has been implicated in cases of cervical intraepithelial neoplasia (CIN) and invasive cervical cancer (ICC). Epidemiological studies indicate that HPV-45 accounts for approximately 4%-8% of cervical cancers, highlighting its relevance in public health discussions on HPV-related diseases. In Europe, HPV-45 has been identified as one of the top three HPV types detected in cervical adenocarcinomas, emphasizing its clinical significance alongside other high-risk types.

The oncogenic mechanisms of HPV-45 are closely associated with its early proteins, particularly E6 and E7. These proteins disrupt critical cellular regulatory pathways by inactivating tumor suppressors such as p53 and retinoblastoma (pRb), resulting in uncontrolled cell proliferation and genomic instability. HPV-45 infection can lead to integration of its genome into host DNA, a process often linked to persistent infection and progression to cervical cancer. In regions with higher prevalence, the association of HPV-45 with cervical adenocarcinoma has become increasingly recognized, highlighting the importance of understanding its interactions with host cellular pathways and the immune system.

Controlling HPV-45 infections through vaccination and screening is essential for improving cervical cancer prevention. Although current vaccines, such as Gardasil and Cervarix, primarily target HPV-16 and HPV-18, evidence suggests some cross-protective efficacy against HPV-45. Implementing comprehensive screening strategies that include HPV-45 can enhance early detection and treatment outcomes. Raising awareness and developing targeted intervention strategies are critical for managing the public health impact of HPV-45, especially in regions where its prevalence is rising and its contribution to malignancy is more pronounced.
