GNRH1 Introduction
GNRH1 exists as gonadotropin-releasing hormone 1 which is a decapeptide neurohormone that the hypothalamus produces mainly from specialized neurons. The prepro-GnRH precursor undergoes enzymatic processing to produce the biologically active GnRH decapeptide and an associated peptide. The vertebrate reproductive axis depends on GnRH1 to function as the primary hypothalamic signal which connects brain signals to pituitary-gonadal system control. The system maintains tight developmental and physiological control over GnRH1 release through neuronal signals and endocrine and peripheral feedback mechanisms to produce the specific pulsatile pattern needed for reproductive development and fertility.

GnRH1 peptide functions by attaching to GnRH receptors found on pituitary gonadotrophs which activates multiple signaling pathways that lead to increased production and release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). The reproductive system requires GnRH1 to release in specific patterns because different signal strengths and frequencies control how LH and FSH genes express themselves and how much they get released. Research indicates GnRH1 performs additional functions beyond reproductive control because it influences behavior and metabolism and shows neuromodulatory effects on the body.
The reproductive system develops disorders because of changes that occur in GNRH1 gene expression and peptide processing and secretion patterns. The GNRH1 gene contains pathogenic mutations which cause normosmic congenital hypogonadotropic hypogonadism (nCHH) leading to delayed or absent puberty and infertility because it fails to activate pituitary gonadotropin production. The GnRH pathway dysfunction leads to three conditions: polycystic ovary syndrome and precocious puberty and specific hormone-dependent cancers which include prostate and breast tumors. Research indicates that this neuroendocrine hormone might link to new neuropsychiatric and metabolic conditions which would increase its clinical importance.
