GALNT6 Full Name
UDP-N-acetyl-alpha-D-galactosamine:polypeptide N-acetylgalactosaminyltransferase 6 (GalNAc-T6)
GALNT6 Introduction
GALNT6, officially designated as UDP-N-acetyl-alpha-D-galactosamine:polypeptide N-acetylgalactosaminyltransferase 6, is a critical member of the large GalNAc-transferase enzyme family. This family is responsible for initiating mucin-type O-glycosylation, a fundamental post-translational modification in metazoan biology. GALNT6 specifically catalyzes the transfer of an N-acetylgalactosamine (GalNAc) residue from the donor sugar nucleotide UDP-GalNAc to the hydroxyl group of serine or threonine residues on target protein substrates. This enzymatic action represents the first and often rate-limiting step in the biosynthesis of complex O-glycan structures, which are pivotal for a myriad of cellular functions, including protein stability, receptor signaling, and cell-cell adhesion.

Structural Architecture and Catalytic Mechanism
The GALNT6 protein features a conserved bi-lobed catalytic domain characteristic of all GalNAc-T isoforms, which includes a tightly bound manganese ion essential for coordinating the UDP-GalNAc sugar donor. A unique feature distinguishing GALNT6 from other family members is the variable loop region within its lectin domain, which confers distinct peptide substrate specificity. This enzyme operates via a sequential, ordered Bi-Bi mechanism where the sugar donor binds first, followed by the acceptor peptide. The subsequent catalytic step involves a nucleophilic attack that transfers the GalNAc moiety, after which the product is released. The presence of a C-terminal ricin-like lectin domain further aids in binding GalNAc-glycosylated peptides, potentially enabling a processive glycosylation mechanism that determines the final glycan density on target proteins.
Pathophysiological Implications and Translational Significance
Dysregulation of GALNT6 expression and activity has been extensively documented in various malignant pathologies, particularly in breast, ovarian, and pancreatic carcinomas, where it often correlates with aggressive tumor phenotypes and poor clinical prognosis. In these contexts, altered O-glycosylation driven by GALNT6 can promote oncogenic processes, including enhanced cell migration, evasion of apoptosis, and resistance to chemotherapeutic agents. This strong association has elevated GALNT6 to a promising candidate for both a diagnostic biomarker and a therapeutic target. Current research is actively exploring small molecule inhibitors and monoclonal antibodies that can specifically block its enzymatic function, aiming to disrupt tumor-promoting glycan signatures while sparing normal tissues, potentially offering a novel anti-cancer strategy that targets the glycosylation machinery directly.
